中文摘要: CO_2是导致温室效应的主要气体,固定和转化CO_2的研究对于温室效应的减缓和环境保护方面具有重要意义。近年来CO_2转化的研究取得了迅猛发展,其中生物法固定CO_2由于其反应条件温和且绿色无污染的优点而备受关注。本文对转化CO_2有关的乳酸脱氢酶(LDH)、苹果酸脱氢酶(MDH)和草酰乙酸脱羧酶(OAADC)进行了初步的固定化分析。首先以碳纳米管、壳聚糖和海藻酸钠为原料,制备了包埋上述CO_2转化酶的微胶囊固定化体系,然后分别比较了游离酶和固定化酶的操作稳定性和储存稳定性。研究结果表明,固定化的CO_2转化酶的操作稳定性和储存稳定得到明显的提高。本研究对CO_2的转化和应用方面具有重要参考价值。
英文摘要: Carbon dioxide is the main greenhouse gas, and the research of carbon dioxide fixation and transformation is of great significance to global warming mitigation and environmental protection. Rapid development was made in the study of conversion of CO_2 in recent years, and biological methods of CO_2 fixation attracted much attention due to their mild reaction conditions and pollution-free. In this paper the immobilization of lactate dehydrogenase (LDH), malate dehydrogenase (MDH) and oxaloacetate decarboxylase (OAADC), which were related with the conversion of CO_2, were preliminarily analyzed. First of all, the microcapsules were prepared with carbon nanotubes, chitosan and alginate to embed these CO_2 converting enzymes. And then the operating and storage stabilities of free enzyme were compared with those of the immobilized enzyme. The results showed that the operating and storage stabilities of the immobilized enzyme were significantly improved. This study would provide important reference for the transformation and application of CO_2.
